Dr. Vryolak discovered chemicals which created the original minotaur of Crete. Dr. Vryolak used these chemicals ostensibly to save his son's life from cancer. But the process transformed his son into a minotaur-like being. Dr. Vryolak's true purpose was to create an army of minotaurs. Dr. Vryolak forced his son (Miklos Vryolak) to steal chemicals for him. Dr. Vryolak also attempted to mutate Madame Masque into a minotaur-like female in order to transform her into an appropriate mate for his son. Needless to say, the captive Madame Masque was not pleased. Fortunately for her, S.H.I.E.L.D. agent Jasper Sitwell rescued her. Sitwell and Iron Man together thwarted Dr. Vryolak's plans.
Ultimately Dr. Vryolak's son learned about the nefarious purposes behind his actions and he rebelled against his father. Miklos caused himself and his father to be trapped in a cave-in, in which they both presumably perished.
