Mulligan was part of a team of scientists and medical personnel who were experimenting on Doctor Octopus. They brainwashed him in order to use him as an assassin to kill Norman Osborn. This was all done at the behest of a shadowy secret cabal that had, since the 1940s, subtly recruited and manipulated super-villains in order to combat the "threat" of super-heroes.
Mulligan probably knew little or nothing about this secret cabal. He may have simply been a lower level scientist or technician.
Norman Osborn knew about the impending attempt on his life, using Doctor Octopus as an assassin. Fearing for his life, he arranged an elaborate scheme in which Mac Gargan (Scorpion) kidnapped Aunt May and blackmailed Spider-Man into helping him (Osborn) escape from Riker's Island Prison.
During the escape, Mulligan rushed into the operating room where Doctor Octopus was being subjected to experiments and brainwashing treatment. Mulligan made the mistake of loudly announcing Osborn's escape. Doctor Octopus heard this and reacted violently. He had already been primed to kill Osborn, so he escaped from the researches and took off, looking for Osborn.
In panel 5 on page 19, one of the other researchers working on Doctor Octopus belated Mulligan, saying: "Just be glad we don't put you on report for this, Mulligan. Coming in here and..."
